摘要:
目的:觀察骨疏康膠囊對去卵巢大鼠骨小梁的影響。方法:將120只6月齡雌性SD大鼠隨機(jī)分為空白組、模型組、骨疏康組和仙靈骨葆組,每組30只。除空白組外,其余3組大鼠均切除雙側(cè)卵巢進(jìn)行骨質(zhì)疏松造模。造模手術(shù)3 d后,骨疏康組、仙靈骨葆組分別以骨疏康膠囊(4.32 g·kg-1·d-1)、仙靈骨葆膠囊(3 g·kg-1·d-1)與蒸餾水配成的藥液灌胃,空白組和模型組均以等量蒸餾水灌胃,共持續(xù)30 d。藥物干預(yù)結(jié)束后處死大鼠,取雙側(cè)股骨制成HE染色切片,測定并比較各組大鼠的股骨骨小梁面積、骨小梁厚度、骨小梁間距及骨小梁面積百分?jǐn)?shù)。結(jié)果:4組大鼠股骨骨小梁厚度、骨小梁間距、骨小梁面積及骨小梁面積百分?jǐn)?shù)比較,組間差異均有統(tǒng)計學(xué)意義[(118.74±8.93)μm,(52.00±6.49)μm,(107.00±9.02)μm,(85.00±4.38)μm,F=5.382,P=0.038;(258.37±78.48)μm,(564.19±135.83)μm,(301.37±89.54)μm,(426.48±98.32)μm,F=7.482,P=0.006;(648 092.35±11 353.46)μm2,(254 872.04±7 429.42)μm2,(589 435.58±10 374.58)μm2,(465 683.69±14 294.49)μm2,F=13.272,P=0.004;(0.49±0.02)%,(0.17±0.01)%,(0.41±0.03)%,(0.29±0.02)%,F=2.143,P=0.019]。模型組的骨小梁厚度、骨小梁面積及骨小梁面積百分?jǐn)?shù)均小于空白組(P=0.002; P=0.001; P=0.014),骨小梁間距大于空白組(P=0.014); 骨疏康組和仙靈骨葆組的骨小梁厚度、骨小梁面積、骨小梁面積百分?jǐn)?shù)均大于模型組(P=0.025; P=0.017; P=0.019; P=0.042; P=0.015; P=0.022),骨小梁間距均小于模型組(P=0.019; P=0.019); 骨疏康組的骨小梁厚度、骨小梁面積、骨小梁面積百分?jǐn)?shù)均大于仙靈骨葆組(P=0.003; P=0.001; P=0.017),骨小梁間距小于仙靈骨葆組(P=0.016)。結(jié)論:骨疏康膠囊能明顯增大去卵巢大鼠骨組織的骨小梁厚度、骨小梁面積及骨小梁面積百分?jǐn)?shù),減小骨小梁間距,其效果優(yōu)于仙靈骨葆膠囊。
Abstract:
Objective:To observe the effect of Gushukang Jiaonang(骨疏康膠囊,GSKJN)on bone trabecula in the ovariectomized rats.Methods:One hundred and twenty 6-month-old female SD rats were randomly divided into blank group,model group,GSKJN group and Xianling Gubao Jiaonang(仙靈骨葆膠囊,XLGBJN)group,30 cases in each group.The rats in model group,GSKJN group and XLGBJN group were administrated with ovariectomy to build models of osteoporosis.Three days after modeling operation,the rats in GSKJN group and XLGBJN group were intragastric administrated with mixed solution of distilled water combined with GSKJN(4.32g/kg/day)and XLGBJN(3g/kg/day)respectively,while the rats in blank group and model group were intragastric administrated with equivalent distilled water for consecutive 30 days.After drug intervention,the rabbits were executed and their bilateral femurs were fetched out and sectioned for HE staining.Then the femoral trabecular area,trabecular thickness,trabecular interval and percentage of trabecular area were measured and compared between the 4 groups.Results:There were statistical differences in femoral trabecular thickness,trabecular interval,trabecular area and percentage of trabecular area between the 4 groups(118.74+/-8.93,52.00+/-6.49,107.00+/-9.02,85.00+/-4.38 μm,F=5.382,P=0.038; 258.37+/-78.48,564.19+/-135.83,301.37+/-89.54,426.48+/-98.32 μm,F=7.482,P=0.006; 648092.35+/-11353.46,254872.04+/-7429.42,589435.58+/-10374.58,465683.69+/-14294.49 μm(2),F=13.272,P=0.004; 0.49+/-0.02,0.17+/-0.01,0.41+/-0.03,0.29+/-0.02%,F=2.143,P=0.019).The trabecular thickness,trabecular area and percentage of trabecular area were less in model group compared with blank group(P=0.002; P=0.001; P=0.014),and the trabecular interval was greater in model group compared with blank group(P=0.014).The trabecular thickness,trabecular area and percentage of trabecular area were greater and the trabecular interval was less in GSKJN group and XLGBJN group compared with model group(P=0.025; P=0.017; P=0.019; P=0.042; P=0.015; P=0.022; P=0.019; P=0.019).The trabecular thickness,trabecular area and percentage of trabecular area were greater and the trabecular interval was less in GSKJN group compared with XLGBJN group(P=0.003; P=0.001; P=0.017; P=0.016).Conclusion:Oral application of GSKJN can obviously increase the trabecular thickness,trabecular area and percentage of trabecular area and decrease the trabecular interval in the ovariectomized rats,moreover,its therapeutic effect is better than that of XLGBJN.
