摘要:
骨折是骨科最常見的一類疾病。疼痛是骨折患者的首發(fā)癥狀,且較劇烈。臨床上常用的止痛藥物——非甾體抗炎藥(non-steroidal anti-inflammatory drug,NSAID)和麻醉性鎮(zhèn)痛藥均能有效緩解骨折疼痛,但是癥狀容易反復(fù),而且上述藥物具有一定程度的不良反應(yīng)。近年來的研究顯示,神經(jīng)生長因子(nerve growth factor,NGF)/原肌球蛋白受體激酶A(tropomyosin receptor kinase A,TRKA)信號通路與骨折疼痛的產(chǎn)生與傳導(dǎo)密切相關(guān),阻斷或抑制該信號通路能明顯緩解骨折疼痛,但該信號通路與骨折愈合的關(guān)系尚不明確。目前針對NGF/TRKA信號通路研發(fā)的藥物表現(xiàn)出較好的緩解骨折疼痛的作用,有望成為臨床上治療骨折疼痛的新選擇。本文就NGF/TRKA信號通路與骨折疼痛和骨折愈合之間關(guān)系的研究進(jìn)展進(jìn)行了綜述。
