摘要:
目的:探討蠲痹歷節(jié)清方對急性痛風(fēng)性關(guān)節(jié)炎大鼠的影響及其作用機制。方法:將96只2月齡雄性清潔級健康SD大鼠隨機分為正常對照組、模型組、蠲痹歷節(jié)清方組、依托考昔組,每組24只。模型組、蠲痹歷節(jié)清方組、依托考昔組大鼠以氧嗪酸鉀溶液連續(xù)腹腔注射后,于大鼠右側(cè)踝關(guān)節(jié)腔內(nèi)注射尿酸鈉溶液; 正常對照組大鼠腹腔及右側(cè)踝關(guān)節(jié)腔分別注入等劑量生理鹽水。關(guān)節(jié)腔內(nèi)注射后即刻給予藥物干預(yù),正常對照組和模型組以10 mL·kg-1生理鹽水灌胃,蠲痹歷節(jié)清方組以22 g·kg-1劑量的蠲痹歷節(jié)清方懸混液灌胃,依托考昔組以5.5 mg·kg-1劑量的依托考昔懸混液灌胃; 每日2次,連續(xù)灌胃2 d。分別于藥物干預(yù)開始后4 h、12 h、24 h、48 h采用免疫組化法檢測各組大鼠踝關(guān)節(jié)滑膜組織中腫瘤壞死因子α(tumor necrosis factor-α,TNF-α)、白細胞介素
Abstract:
ABSTRACT Objective:To explore the effects of Juanbi Lijie Qing Fang(蠲痹歷節(jié)清方,JBLJQF)on rats with acute gouty arthritis and its mechanism of action.Methods:Ninety-six 2-month-old healthy clean-grade male SD rats were randomly divided into normal control group,model group,JBLJQF group and etoricoxib group,24 cases in each group.The rats in model group,JBLJQF group and etoricoxib group were intraperitoneal injected continuously with oteracil potassium solutions,and then the sodium urate solutions were injected into their right ankles,while the rats in normal control group were administrated with intraperitoneal injection and intra-articular injection of the same dose of normal saline(NS)respectively.Drug interventions were performed on the rats immediately after intra-articular injection.The rats in normal control group,model group,JBLJQF group and etoricoxib group were intragastric administrated with NS(10 mL/kg),NS(10 mL/kg),JBLJQF suspension(22 g/kg)and etoricoxib suspension(5.5 mg/kg)respectively,twice a day for 2 consecutive days.The contents of tumor necrosis factor-α(TNF-α)protein,interleukin-1β(IL-1β)protein and interleukin-6(IL-6)protein in ankle synovial tissues were detected by using immunohistochemical method at 4,12,24 and 48 hours after the beginning of drug intervention respectively.The blood uric acid(UA)contents were detected by using full-
