摘要:
目的:觀察強骨飲對去卵巢骨質疏松大鼠骨顯微結構的影響。方法:將36只10周齡雌性SD大鼠隨機分為假手術組、強骨飲組和模型組,每組12只。假手術組切除卵巢周圍脂肪,其他兩組均切除雙側卵巢。造模后3個月,假手術組以生理鹽水0.041 mL·g-1灌胃; 強骨飲組以強骨飲濃縮液0.041 mL·g-1灌胃; 模型組以碳酸鈣D3混懸液0.01 mL·g-1、阿法骨化醇溶液0.037 mL·g-1灌胃; 每日1次,連續(xù)干預2個月。藥物干預結束后,采用micro-CT掃描大鼠股骨頭至股骨中段,對圖像進行三維重建,觀察大鼠骨顯微結構變化,并對3組大鼠各項骨顯微結構參數(shù)進行比較。結果:藥物干預2個月后,假手術組骨組織內骨小梁結構排列緊密、有序,骨小梁分離度低; 強骨飲組和模型組骨小梁數(shù)量明顯減少,排列稀疏或出現(xiàn)斷裂,形態(tài)扭曲。3組間骨表面積(bone surface,BS)、骨體積(bone volume,BV)、骨表面積和體積比值(bone surface/bone volume,BS/BV)、骨體積分數(shù)(bone volume/tissue volume,BV/TV)、骨小梁厚度(trabecular thickness,Tb.Th)、骨小梁數(shù)量(trabecular number,Tb.N)、骨小梁分離度(trabecular separation,Tb.Sp)比較,差異均有統(tǒng)計學意義(F=63.243,P=0.000; F=10.290,P=0.000; F=43.774,P=0.000; F=10.685,P=0.044; F=10.489,P=0.000; F=140.827,P=0.000; F=382.812,P=0.000); 強骨飲組和模型組BS、BV、BS/BV、BV/TV、Tb.Th、Tb.N均小于假手術組(P=0.000,P=0.018,P=0.000,P=0.045,P=0.004,P=0.037; P=0.000,P=0.008,P=0.000,P=0.031,P=0.004,P=0.000),Tb.Sp均大于假手術組(P=0.000; P=0.000); 強骨飲組BS、BV、BS/BV、Tb.Th大于模型組(P=0.009,P=0.007,P=0.045,P=0.000),Tb.Sp小于模型組(P=0.000),2組BV/TV和Tb.N的組間差異均無統(tǒng)計學意義(P=0.104,P=0.869)。結論:與碳酸鈣D3和阿法骨化醇聯(lián)用相比,強骨飲更有利于改善去卵巢骨質疏松大鼠的骨顯微結構,具有更好的抗骨質疏松作用。
Abstract:
Objective:To observe the effect of Qianggu Yin(強骨飲,QGY)on bone microstructure in ovariectomized osteoporosis rats.Methods:Thirty-six 10-week-old female SD rats were randomly divided into sham-operated group,QGY group and model group,12 cases in each group.The lipectomy around the ovary were performed on rats in sham-operated group,while the bilateral ovariectomy were performed on rats in QGY group and model group.The rats in sham-operated group were intragastric administrated with normal saline(0.041 ml/g),and the rats in QGY group were intragastric administrated with QGY concentrated solution(0.041 ml/g),while the others in model group were intragastric administrated with calcium carbonate D3 suspensions(0.01ml/g)and alfacalcidol solution(0.037ml/g)at 3 months after the modeling,once a day for consecutive 2 months.After the end of drug intervention,the femoral head and middle segment of femur of the rats were scaned by micro-CT,and the images were inputed into the computer to build three-dimensional reconstructed images.Then the bone microstructure changes in rats were observed and the bone microstructure parameters were compared between the 3 groups.Results:After 2-month drug intervention,compact and orderly bone trabecula were found in the bone tissue in sham-operated group with low separation degree,while sparse or broken bone trabecula were found in QGY group and model group with distorted shape.There was statistical difference in bone surface(BS),bone volume(BV),ratio of bone surface to bone volume(BS/BV),bone volume fraction(BV/TV),trabecular thickness(Tb.Th),trabecular number(Tb.N)and trabecular separation(Tb.Sp)between the 3 groups(F=63.243,P=0.000; F=10.290,P=0.000; F=43.774,P=0.000; F=10.685,P=0.044; F=10.489,P=0.000; F=140.827,P=0.000; F=382.812,P=0.000).BS,BV,BS/BV,BV/TV,Tb.Th and Tb.N were less and Tb.Sp were greater in QGY group and model group compared to sham-operated group(P=0.000,P=0.018,P=0.000,P=0.045,P=0.004,P=0.037; P=0.000,P=0.008,P=0.000,P=0.031,P=0.004,P=0.000; P=0.000; P=0.000).BS,BV,BS/BV and Tb.Th were greater and Tb.Sp were less in QGY group compared to model group(P=0.009,P=0.007,P=0.045,P=0.000,P=0.000),and there were no statistical difference in BV/TV and Tb.N between the 2 groups(P=0.104,P=0.869).Conclusion:QGY is more conducive to improving the bone microstructure in ovariectomized osteoporosis rats and it has better anti-osteoporotic effect compared to the combination therapy of calcium carbonate D3 and alfacalcidol.
