摘要:
目的:探討動(dòng)態(tài)溫?zé)崾杳懿▽切躁P(guān)節(jié)炎軟骨細(xì)胞凋亡調(diào)控基因表達(dá)的影響。方法:大白兔60只,隨機(jī)分為2組,即:正常組(10只)和手術(shù)組(50只)。手術(shù)組采用改良Hulth法復(fù)制膝骨性關(guān)節(jié)炎模型,術(shù)后隨機(jī)分為5組,模型組造模后常規(guī)飼養(yǎng),對照組微波治療儀,實(shí)驗(yàn)1組電(疏密波)治療,實(shí)驗(yàn)2組熱(熱軟膜)治療,實(shí)驗(yàn)3組電熱(治療型護(hù)膝動(dòng)態(tài)溫?zé)崾杳懿?治療,分別對應(yīng)治療16周后,觀察關(guān)節(jié)軟骨的組織形態(tài)學(xué),檢測軟骨細(xì)胞的凋亡率及p21、p53、Bcl-2 mRNA的表達(dá)。結(jié)果:16周時(shí),骨性關(guān)節(jié)炎模型成功建立,經(jīng)治療后各組關(guān)節(jié)軟骨組織形態(tài)學(xué)病理變化程度明顯輕于模型組(P<0.01,P<0.05); 各組軟骨細(xì)胞的凋亡率、p53 mRNA表達(dá),模型組、對照組、實(shí)驗(yàn)1組、實(shí)驗(yàn)2組、實(shí)驗(yàn)3組明顯高于正常組(P<0.01),對照組、實(shí)驗(yàn)2組、實(shí)驗(yàn)3組明顯低于模型組、實(shí)驗(yàn)1組(P<0.01,P<0.05),實(shí)驗(yàn)3組明顯低于對照組、實(shí)驗(yàn)2組(P<0.01,P<0.05); 軟骨細(xì)胞p21、Bcl-2 mRNA表達(dá),模型組、對照組、實(shí)驗(yàn)1組、實(shí)驗(yàn)2組、實(shí)驗(yàn)3組明顯低于正常組(P<0.01),對照組、實(shí)驗(yàn)2組、實(shí)驗(yàn)3組明顯高于模型組、實(shí)驗(yàn)1組(P<0.01,P<0.05),實(shí)驗(yàn)3組明顯高于對照組、實(shí)驗(yàn)2組(P<0.01,P<0.05)。結(jié)論:動(dòng)態(tài)溫?zé)崾杳懿苡行抡{(diào)軟骨細(xì)胞p53 mRNA表達(dá),上調(diào)p21、Bcl-2 mRNA表達(dá),從而抑制軟骨細(xì)胞的凋亡,延緩關(guān)節(jié)軟骨退變。
Abstract:
Objective:To investigate the effect of dynamic warm sparse-dense wave on apoptosis regulatory genes expression of chondrocyte.Methods:60 white rabbits were randomly divided into normal group(n=10)and operation group(n=50).The operation group copied knee osteoarthritis by modified Hulth Law were randomly divided into 5 groups.Model group received conventional breeding after modeled,control group received the microwave therapy,experimental group 1 received electricity(sparse-dense wave)therapy,experimental group 2 received thermal(thermal soft film)therapy,and experimental group 3 received electricity and thermal(dynamic warm sparse-dense wave)therapy.After treatment 16 weeks,the morphology of articular cartilage was observed,apoptosis rate and p21,p53,Bcl-2 mRNA expression of chondrocyte was detected.Results:After 16 weeks,knee osteoarthritis model was successfully established,articular cartilage morphology in each treatment group level of pathological changes significantly reduced than its in model group(P<0.01,P<0.05).Apoptosis ate and p53 mRNA expression of chondrocyte,model group,control group,experimental group 1,experimental group 2 and experimental group 3 was significantly higher than the normal group(P<0.01),control group,experimental group 2 and experimental group 3 was significantly lower than the model group,experimental group 1(P<0.01,P<0.05),experimental group 3 was significantly lower than control group,experimental group 2(P<0.01,P<0.05); The expression of p21、Bcl-2 mRNA, Model group,control group,experimental group 1,experimental group 2,experimental group 3 was significantly lower than normal group(P<0.01),control group,experimental group 2 and experimental group 3 was significantly higher than model group,experimental group 1(P<0.01,P<0.05),experimental group 3 was significantly higher than control group,experimental group 2(P<0.01,P<0.05).Conclusion:Dynamic warm sparse-dense wave can be effect of down-regulating the expression of p53 mRNA,up-regulating the expression of p21、Bcl-2 mRNA,inhibit chondrocyte apoptosis and delay the degeneration of cartilage.
