摘要:
目的:探討受體酪氨酸激酶抑制劑genistein對(duì)兔OA模型軟骨細(xì)胞NO的表達(dá)及軟骨組織形態(tài)的影響,并探討其可能的作用機(jī)制。方法:參照Hulth法建立兔骨關(guān)節(jié)炎模型,予以genistein干預(yù)后,分別按照0.1 mg·kg-1、0.2 mg·kg-1、0.3 mg·kg-1、0.4 mg·kg-1、0.5 mg·kg-1的給藥劑量進(jìn)行分組,行手術(shù)側(cè)關(guān)節(jié)腔內(nèi)注射,通過(guò)測(cè)定各組亞硝酸鹽含量檢測(cè)關(guān)節(jié)腔內(nèi)NO的表達(dá)水平,同時(shí)進(jìn)行軟骨組織形態(tài)學(xué)檢查。結(jié)果:0.3 mg·kg-1、0.4 mg·kg-1、0.5 mg·kg-1濃度genistein干預(yù)組關(guān)節(jié)腔內(nèi)NO的表達(dá)水平明顯低于對(duì)照組,差異具有統(tǒng)計(jì)學(xué)意義(P<0.05); 對(duì)軟骨組織行H.E/SOFG染色顯示,在造模后第4周、第8周與第12周時(shí),genistein干預(yù)組與對(duì)照組相比,組織損傷均明顯減輕,Mankin評(píng)分均明顯下降,差異具有統(tǒng)計(jì)學(xué)意義(P<0.05)。結(jié)論:受體酪氨酸激酶抑制劑genistein在體內(nèi)能減少骨關(guān)節(jié)炎軟骨細(xì)胞NO的表達(dá),對(duì)于延遲關(guān)節(jié)軟骨在組織形態(tài)水平的退變是有效的, 但是尚不能最終阻止軟骨退變發(fā)生發(fā)展的進(jìn)程。
Abstract:
Objective:To investigate the effects of recePtor tyrosine kinase inhibitor genistein on the exPression of NO and ultra-microstructure of osteoarthritis cartilage and chondrocyte in OA rats, thus try to exPress the Probable mechanisms involved.Methods:According to Hulth method,we built rat OA model and the exPression levels of NO was detected by Griess method and the change of ultra-microstructure of cartilage was observed.Results:ComPared with control grouP, less levels of NO were detected after the intervention of genistein(0.3 mg·kg-1、0.4 mg·kg-1 and 0.5 mg·kg-1)(P<0.05). At 4、8 and 12 weeks after oPeration, cartilage sections made and stain with HE/SOFG stain showed less injury, and Mankin score were significantly decreased in intervention grouPs comPared with control grouP(P<0.05).Conclusion:Genistein can reduced the exPression of NO in-vivo. The results of Mankin score and HE/SOFG stain showed that genistein Partly delayed the observation of ultra-microstructure of cartilage exPlored the alleviation of the ultra-microstructure damage of cartilage after injury.
