摘要:
目的:探討復(fù)元活血湯含藥血清對大鼠成骨細(xì)胞功能的影響及其作用機(jī)制。方法:取2月齡SD雄性大鼠12只,隨機(jī)分為2組, 每組6只,分別給予20 g·kg-1復(fù)元活血湯生藥灌胃(含藥血清組)和同等量的生理鹽水灌胃(空白血清組),每天2 次,14 d后腹主動脈采血。取新生SD大鼠2只,采用二次酶消化法消化幼鼠顱蓋骨獲取成骨細(xì)胞,采用血清藥理學(xué)方法,分別采用含 胎牛血清的培養(yǎng)液(胎牛血清組)、含復(fù)元活血湯生藥灌胃大鼠血清的培養(yǎng)液(含藥血清組)和含生理鹽水灌胃大鼠血清的培養(yǎng)液( 空白血清組)培養(yǎng)成骨細(xì)胞。分期測定體外培養(yǎng)的大鼠成骨細(xì)胞的增殖功能、分化功能、礦化功能及堿性磷酸酶含量。結(jié)果:① 成骨細(xì)胞增殖功能。時(shí)間效應(yīng)F=381.260,P=0.000; 組別效應(yīng)F=1 071.170,P=0.000; 時(shí)間與組別交互效應(yīng)F=22.500,P=0.000。 ②成骨細(xì)胞分化功能。時(shí)間效應(yīng)F=159.950,P=0.000; 組別效應(yīng)F=62.120,P=0.000; 時(shí)間與組別交互效應(yīng)F=2.400,P=0.029。③ 成骨細(xì)胞礦化功能。時(shí)間效應(yīng)F=115.500,P=0.000; 組別效應(yīng)F=4.370,P=0.082; 時(shí)間與組別交互效應(yīng)F=56.300,P=0.000。④成 骨細(xì)胞堿性磷酸酶含量。時(shí)間效應(yīng)F=2 175.670,P=0.000; 組別效應(yīng)F=1 201.370,P=0.000; 時(shí)間與組別交互效應(yīng) F=428.870,P=0.000。結(jié)論:復(fù)元活血湯能明顯地促成骨細(xì)胞的增殖和分化功能,其作用可能與成骨細(xì)胞合成和分泌堿性磷酸酶有 關(guān)。
Abstract:
Objective:To study the effect of serum containing FuYuanHuoXue Recipe on the function of rat's osteoblasts in vitro and the mechanism of action.Methods:12 SD maleness rats aged 2 months were divided into two groups with 6 rats in each group at random,rats in one group(experimental group)undergone intragastric administration of FuYuanHuoXue decoction(20 g·kg-1)while equivalent normal saline was administrated in another group(control group)twice everyday,blood was obtained from abdominal aorta 14 days later.Osteoblasts were obtained from 2 neonatal rats and cultured respectively with fluid containing fetal bovine serum,serum from the rats in experimental group and control group.Function of osteoblasts was evaluated according to the measurement of proliferation,differentiation,mineralization and quantity of alkaline phosphatase(AKP).Results: ①Proliferation of osteoblast:time effect:F=381.260,P=0.000; effect between groups:F=1 071.170,P=0.000; reciprocal effect between time and group:F=22.500,P=0.000.②Differentiation of osteoblast:time effect:F=159.950,P=0.000; effect between groups:F=62.120,P=0.000; reciprocal effect between time and group:F=2.400,P=0.029.③Mineralization of osteoblast:time effect:F=115.500,P=0.000; effect between groups:F=4.370,P=0.082; reciprocal effect between time and group:F=56.300,P=0.000.④content of AKP:time effect:F=2 175.670,P=0.000; effect between groups:F=1 201.370,P=0.000; reciprocal effect between time and group:F=428.870,P=0.000.Conclusion:FuYuanHuoXue decoction can incite proliferation and differentiation of rat's osteoblast in vitro owing to the promotion in secretion of alkaline phosphatase.
